Description
This session will review the basic scientific principles governing the successful cryopreservation of current cellular therapy/transfusion medicine products. A focus will be on limits that cell biophysics, ice crystal nucleation and growth, cryoprotectant toxicity and water and cryoprotectant permeability have on the development of a successful cryopreservation protocol. The session will be delivered using a "standard" cryopreservation protocol as the backdrop from which the basic scientific principles will be discussed. Furthermore, multiple cryopreservation freezing curve results that led to positive and negative cellular therapy product results will be presented. Analysis and interpretation of the data, failure analysis of the cryopreservation issues and clinical relevance will be offered. This session is related to a project of the Product Manufacturing and Testing (PMAT) Subsection of the AABB Cellular Therapy Section.

  • Present data from multiple clinically related cryopreservation freezing curve issues and discuss analysis/interpretations of the freezing curve issues and clinical relevance.
  • Provide a summary of the key advances in cryobiology that resulted in the current cellular therapy and transfusion medicine cryopreservation protocols.
  • Summarize the scientific basis for the critical steps in clinical cryopreservation protocols to provide end users with a more complete understanding of the need to consider many factors when designing, validating and implementing a cryopreservation protocol.
